Article Title: Cu 2+ -chelated epigallocatechin gallate nanoparticle-functionalized hydrogel promotes osteogenesis by inhibiting ferroptosis.
Article Snippet: A pathological inflammatory microenvironment can hinder bone regeneration by disrupting the osteogenic differentiation potential of bone marrow-derived mesenchymal stem cells (BMSCs).. However, conventional antiinflammatory therapies alone are insufficient to fully resolve this challenge.. Bone defect repair is often impaired by persistent inflammation-induced ferroptosis in BMSCs, and traditional approaches fail to prevent lipid peroxidation-driven cell death.

Article Title: Increased extracellular fluid viscosity drives M2-like macrophage polarization to promote breast cancer progression and immunotherapy resistance.
Article Snippet: .. For IHC staining, tumor tissues were incubated overnight at 4◦C with the following primary antibodies, anti-F4/80 (1:200, #ab90247, Abcam) and anti-CD206 (1:10000, #18704-1-AP, proteintech). ..

Article Title: METTL1-Mediated m7G Modification of GINS2 mRNA Activates STAT3 to Drive Esophageal Squamous Cell Carcinoma Progression.
Article Snippet: Background: METTL1mediated m7G modification plays a procancer role in various malignant tumors, but whether it promotes the progression of esophageal squamous cell carcinoma (ESCC) by regulating the DNA replication factor GINS2 and tumorassociated macrophage polarization remains unclear.. Methods: The expression and function of GINS2 were examined through in vitro and in vivo experiments.. RIP, MeRIP, and RNA stability assays were used to validate the regulatory effect of METTL1 on GINS2 modification.

Article Title: Legumain Restrains Granuloma Formation by Inhibiting mTORC1/STAT1-Mediated M1 Macrophage Polarization in Sarcoidosis.
Article Snippet: After mixing with loading buffer and heating at 100◦C for 10 min, 30–50 μg of protein was separated by 10% sodium dodecyl sulfate (SDS)–polyacrylamide gel electrophoresis and subsequently transferred to polyvinylidene difluoride (PVDF) membranes (Millipore, Billerica, MA, USA). .. The membranes were blocked with 5% skimmed milk for 2 h and incubated overnight at 4◦C with the following primary antibodies: anti-LGMN (Abcam, Cambridge, UK), anti-iNOS (Abcam, Cambridge, UK), anti-Arg1 (Proteintech, Wuhan, China), anti-CD206 (Proteintech, Wuhan, China), anti-STAT1 (Abcam, Cambridge, UK), anti-pSTAT1(Tyr701) (Cell Signaling Technology, Danvers, MA, USA), anti-pSTAT1(Ser727) (Cell Signaling Technology, Danvers, MA, USA), anti-JAK1 (Proteintech, Wuhan, China), anti-pJAK1(Tyr1034/1035) (Cell Signaling Technology, Danvers, MA, USA), anti-ITGAV (Abcam, Cambridge, UK), anti-ITGB3 (Abcam, Cambridge, UK), anti-pS6 (Cell Signaling Technology, Danvers, MA, USA), anti-S6 (Cell Signaling Technology, Danvers, MA, USA), anti-HK2 (Cell Signaling Technology, Danvers, MA, USA), anti-LDHA (Proteintech, Wuhan, China), and anti- β-actin (Proteintech, Wuhan, China). .. For M1 polarization, BMDMs were treated ith 50 ng/mL LPS (Sigma-Aldrich, Merck KGaA, Darmstadt, ermany) and 20 ng/mL recombinant mouse IFN- γ (PeproTech, ranbury, NJ, USA) and harvested at the indicated time points 0, 3, 6, and 12 h) for time-course experiments, or at 12 h for all ther experiments.

Article Title: Core fucosylation of NCEH1 by FUT8 promotes progression of high-grade serous ovarian cancer by driving tumor-associated macrophage M2 polarization.
Article Snippet: High-grade serous ovarian cancer (HGSC) is the most aggressive subtype of ovarian epithelial cancer (OEC), with characters of latestage diagnosis, high recurrence rate, and poor survival outcomes.. Fucosyltransferase 8 (FUT8) is responsible for α1,6-core fucosylation biosynthesis, and aberrant FUT8/α1,6-core fucosylation level is involved in tumor progression.. However, the roles and mechanisms of protein FUT8 and α1,6-core fucosylation in HGSC tumorigenesis and progression remain elusive.

Article Title: Pomegranate-inspired bionic microgel/periosteum composite harnessing reactive oxygen species for immune-engineered bone regeneration.
Article Snippet: Excessive reactive oxygen species (ROS) and unresolved inflammation create a hostile microenvironment that compromises regeneration in critical-sized bone defects.. Here, we developed a pomegranate-inspired bionic microgel/periosteum composite (PBC) featuring a hierarchical “membrane–seed” modular design, in which a periosteum-mimetic PDA/PLLA membrane provided not only ROS counteraction but also a continuous protective interface and defect compartmentalization.. Conductive GelMA/PEDOT:PSS microgels serving as the “seeds” enabled enhanced and prolonged ROS clearance as well as conformal filling of irregular geometries, and increased cell–material contact.

IL-1α, TNF-α, IL-6, TLR9, MyD88, STING, and cGAS levels were normalized to the β-actin level in each sample. p-NF-κB, p-TBK1, and p-IRF3 levels were normalized to their respective total protein levels (NF-κB, TBK1, and IRF3) in each sample. These normalized values were then used for statistical analysis (B, D, F), n = 6. (G) RBC-MB treatment significantly attenuated CpG DNA-induced upregulation of CD86 in macrophages. RAW264.7 cells were treated with 1 μg mL -1 CpG DNA for 24 h, followed by incubation with 0.5 mL RBC-MBs (concentration: 1 × 10 8 particles mL -1 ) for 24 h, then immunostained with an anti-CD86 antibody (green) and DAPI (blue, nuclei), and imaged by fluorescence microscopy. Representative images (left panel) and quantitative fluorescence graph (right panel) are shown. Scale bar: 50 μm, n = 6. (H-J) RBC-MB treatment significantly inhibited hepatic IRI-induced polarization of macrophages toward the M1 phenotype (H) and activation of the TLR9-MyD88 and cGAS-STING pathways (I-J). MASLD rats underwent 45 min of hepatic ischemia followed by reperfusion. At 0, 6, 12, 18, and 24 h post-reperfusion, the rats were treated with either Con-MBs or RBC-MBs. Primary macrophages were isolated from rat livers, then co-stained with CD68 and CD163 followed by flow cytometric analysis (H), n = 3/4, or subjected to western blotting (I-J). CD206, iNOS, TLR9, MyD88, STING, and cGAS levels were normalized to the β-actin level in each sample, and these normalized values were used for statistical analysis (J), n = 3. For (B, D, F, G right panel , H right panel , J), error bars represent mean ± standard deviation. P values were calculated using the unpaired Student's t-test (B, D, F, G right panel ) or one-way analysis of variance (ANOVA) (H right panel , J). *, P < 0.05; **, P < 0.01; ***, P < 0.001; ****, P < 0.0001; ns, not significant.(For interpretation of the references to color in this figure legend, the reader is referred to the Web version of this article.)
